If your server is properly configured, both should work. You should contact your hosting provider about having index.php added as a default file for the webserver.

I already have index.php as default! I want to change it in: forum/ .

I have already written to my hosting provider and it says to contact you, then I opened a ticket from you and you told me to ask on this forum.

A default installation doesn't need index.php to work. I am not sure why yours does. Did you edit the navigation tabs from their originals?

Your post is a little confusing, did you mean you want to move to domain/forum/ ?

If thats the case then you need to create a /forum folder, and move your forum into it.

My homepage has always been domain/forum/

Since I installed the SEO, Has become an index.php.

So, to have domain/forum/, do I have to move the whole forum "Upload"? Or just the forum.php file?

You'll need to talk to the people at DBSEO then. What you're asking for is the default state of a vBulletin 4 installation.
