I need to automatically remove all staff (in one group) from an additional usergroup. Is this possible using the promotions system?

Only if they all meet the Promotion criteria of Reputation/Posts/Days Registered. They would all need to have more of one of those (or less reputation) than anyone else in the usergroup in order to get 'promoted' to another usergroup.
